My husband and I spent 5 days in Orvieto with a car to tour the Tuscany area. The hilltowns took quite awhile to get to and if we did it again would spend two days or maybe 3 at the most to go out and tour then find another town. We had to park our car at the edge of town which took awhile to walk to in the mornings. We got on the Autostrada just to save time. We stayed at an Agriturismo for one night but had a hard time finding a place for dinner. I would stay in a town close to the train station if you won't have a car and close to a restaurant/bar. Siena worked for us last month. Stayed there 4 days and did day trips to Assisi, Pienza and Montepulciano
Stayed at Palazzo Bruchi B and B, wonderful, central, great hosts and access to a kitchen and frig.
Tuscany is really quite large. Also difficult to navigate by train or bus. So a rental car is the best to have. I would look for a base at an agriturismo somewhere near Siena. From there you can explore the Chianti area and also the areas to the south and west.
We are planning a stay at Il Canto del Sole,a B & B about 15 minutes outside of Siena. It gets great reviews on www.tripadvisor.com. website is www.ilcantodelsole.com.
to me Siena is the best by public transit but not really hilly. so far I have stayed in Siena, Cortona, Orvieto,& Assisi (Umbria). if you rent a car you would have more options.
