Does anyone have recommendations on Tuscan vineyards/wine tours that can accommodate a large group (20-30 guests) that are open in May?

All of the high-end wineries require advance reservations and pre-payment. There is no reason why, far enough in advance, you could not book what you want (and make the substantial payment required.)
I will say that the most massive places, that would have the easiest accommodation of your request, might not be my own first choices. We were looking forward to the James-Bond-Movie-Set architecture of Antinori, but the massive size, vulgar crowds at the tasting bar, and impersonal nature of the place was a disappointment. And we had pre-booked for lunch at their substantial restaurant.
What I haven't told you is ... whom to call. At a restaurant in the U.S., it's the Director of Sales. At an Italian winery, I don't know.
It is not nearly as big, and barely in Tuscany, but we enjoyed Avignonesi a lot more. There were almost 20 people on our tour. But their wines are expensive.
