Spending 3 nights in Positano in October.Can you suggest tours of the area? Unsure if to go by car or boat due to weather.We plan on visiting Pompeii when we arrive in Naples by train on our way to Positano.

With 3 nights you have no time for tours.
1 Day take the boat ride for the day to Capri
1 Day take the SITA bus from Positano to Amalfi explore Amalfi, from there take the SITA bus to Ravello, explore Ravello come back and explore Positano if you still have time.
In your in between time and half day on departing or arrival spend exploring the town of Positano.
Don't think you have time for the Path of the Gods or any such hikes and will be tight squeezing in above and enough time to see the town of Positano itself.
I skipped Sorrento but if you get a rainy day and/or ferry to Capri is not running ; you could skip Capri and spend the day in Sorrento (reachable by the same SITA bus that goes to Amlafi, opposite direction).

Having a car in Positano will be more convenient in some ways, but parking could be expensive and in short supply. You'll also have to deal with the usual caveats, including the compulsory International Driver's Permit for each driver listed on the rental form, and avoiding the dreaded ZTL (limited traffic) areas and similar pitfalls.
