We will be travelling via rental car from Piran (near Trieste) to Varenna. GoogleMaps says it's a 5 hour drive. We won't be in any rush to get to Varenna. Are there any spots along the way that we should stop at? Verona perhaps?

Considering you will have very limited time since the drive is long (make it 6 hours), you could choose any of the following along the itinerary:
Padua
Vicenza
Verona
Sirmione
Bergamo.
Any of them would make a nice 3 to 5 hour visit. Leave early so that you can time that visit around lunch time.
