Travelling from Florence to Puglia

Least stressful way to travel to Puglia from Florence, then return to Rome for flight?
Have 5 days after leaving Florence and want to see Matera and Alberobello.
Fly and rent car along the way to visit various sites? Not wanting any prop plane
flights.

It's a very long trip; by train, it takes the better part of a day. Service is better via Bologna, but Bologna to Bari is very long. By car is also very long (more than eight hours driving). The best thing would be flying and renting a car in Bari as both Matera and Alberobello are not easily reached by train. Pisa has more flights than Florence - on Ryanair, that does not fly propeller planes but has its own quirks. But you can't be too choosy as you do not have many options.
