It would help to have additional details of your trip...length of trip, other cities you're covering, where else you've been in Europe.
For a pure transfer, you could do it in one long 10 hour day with 1 or 2 changes. You could also break it up with a few nights in Venice. Six hours to Venice and then Venice to Milan by train in as little as about 2.5 hours.
If you want to take the train, and you want to stop over somewhere and you've never been to Venice, I would stop there. But you should stay for longer than one night.
Other easy stops along the train line include Bolzano (gateway to the Dolomites) and Verona (a usual train connection point and well worth an overnight).
Another possible train route is via Zurich - slightly longer travel time and probably more expensive, but with only one train connection. How to Look Up Train Schedules and Routes Online gives you the DB train schedule link and tips for using it.
