There is a town Amalfi on the coast-do you mean there or on “Amalfi Coast”
Rome to the town Amalfi. Late June wedding.
Probably the easiest would be a fast train to Salerno, then a private driver or taxi service to Amalfi.
Roma Termini train to Salerno and a Travelmar ferry. Easy short walk from the station to the port.
If hiring a driver book from Napoli Centrale or Afragola [driver choice]; equidistant from Salerno and a shorter train ride.
"Mobility challenged"--- does that mean cannot walk more than a 100 yds or is wheelchair bound?
Can you travel with other guests/family to help with navigating various modes of transport needed to get from Rome to your destination?
Will you be heading from the airport directly to "Amalfi" or will you been in Rome for a few days before heading to the wedding venue?
The 'best way' is likely a car service, but expect a fee in the $800-1000 range. (Rome to "Amalfi")
Can you manage taking a train from Rome to either Salerno or Naples and taking a car service from there? This would reduce the cost significantly.

Mobility challenged means my wife uses a cane or walker and is very slow. I'll be lugging all the luggage. I'm in good shape for 75 but I am 75. "One's got to know your limitations."
We took the train from Rome to Salerno for a couple of days and then a Travelmar ferry to the town of Amalfi. You probably will want a taxi to take you from the Salerno train station to the ferry dock, although they are walking distance. (Update: yes, with your luggage, definitely the taxi.)
The town of Amalfi is a steep hill after the main piazza. Be sure to reserve a place right away because there aren’t too many choices directly off of the piazza.. Cars & motorcycles buzz up the main road through town, so be careful navigating around the streets.
Just saw your update reply. You might want to look at some YouTube videos to have a good idea of the town. While you are there, be sure to take a bus or taxi up to Ravello. The views are gorgeous, and the flowers & gardens were spectacular and peaceful at the Villa Rufolo. My daughter & I sat on a bench overlooking the view and just enjoyed the atmosphere! That was in June just as they opened in the morning. The bus/taxi stand to the villa is flat walking, and there is a small cafe at the little piazza.
Positano is very hilly.
Sounds like the train is the obvious choice to Salerno. Trains leave directly from Termini so we won't have to go the main terminal. The less scrambling around the better. We have a room in Amalfi already booked. Thanks
My wife is too mobility challenged, and we have a personal scooter she rides on vacations. With it, we've been to Las Vegas and twice to Europe in the last year.
She rides through airports and right up to the door of the airplane. The scooter is returned to us at the plane door when we arrive at our next stop.
There are limitations on rough cobblestone streets and in places like Venice with all the bridges to go up steps. But I'm capable of picking a 48 lb. scooter on and off taxi's, trains, buses, etc.
I've not made it to the Amalfi Coast, however it appears to be a tough place for anyone with mobility issues to go.
We seem to develop more physical limitations every year so you are among friends here!

Keep your luggage LIGHT and small.
We now travel with 2 RS convertible carry on backpacks. Fits an amazing amount of stuff. Easier to get on/off trains. We also bring a 21" roll on w/flight bag. That works extremely well for us, keeps some hands free.
Take taxis to/from airport/hotels/stations.
Allow more time than you think you need to get to a train. As long as you can walk, you’ll get there. I need to go slow myself.
Always keep the transportation bit simple, it’s worth any additional expense.

Termini will be fine. No stairs.
As I recall Salerno did have stairs.
Napoli Centrale- no stairs. Could get driver from here to Amalfi.
You’d have more time to get off the train in Naples than you would in Salerno..I think?
