Travel from Rome to Montepulciano

Still have the train. Mostly Regionale trains, some R. Velose. Depending on time and route and train two to three hours with at least one change. Are you asking if there is another way? Not sure what you are asking.

I've always taken the train to Chiusi and then a bus to Montepulciano. Bus departs from in front of the train station. Buses run less frequently on Sundays.

Please note that the Montepulciano station is some 9 kms. (6 miles) distant from the town - I would not bet in frequent buses linking the station to town. Getting down in Chiusi and taking bus there looks a more likely solution.
