We four adults are flying in to Madrid at 8:30 AM on 3/31, then need to be in Vernazza that evening. We'll stay there a few days, then head to Florence, then return to Madrid on Saturday 4/7 before flying out the next morning. I haven't really researched how to get between these places (and I'm sure it's been answered here somewhere) and could use your suggestions! I don't think we want a car, if that's possible to avoid.

I'm sure there must have been a really compelling reason why you would have chosen such an, um, unconventional city to fly in and out of, since the bulk of your time is in Italy. If you need to get there that night, then forget a car rental. It's over 1500 km and 15 hrs driving time. Fly into Pisa from Madrid and take the train from there. Fly out of Florence to return to Madrid.
Are you really flying into Madrid? Or did you mean Milan?
You do realize they are around 900 miles apart.
Your best bet is to fly Madrid-Pisa
Then train to Vernazza.
Check Ryanair, Easyjet and Vueling for flight times.
Driving would take between 14-15 hrs.

if it is Milan,as suggested above, take the Malapensa express to Milan Central then Milan central to Vernazza. Use Trenitalia.com for times and prices.
Anne, I have to ask why you're flying in and out of Madrid if you're not going to spend any time there? Why didn't you chose to fly in/out of Italian airports much closer to where you intend to be? You are going to expend a lot of time in transit between Spain and Italy.
If you can't change your flights, flying from Madrid to Pisa, Genoa or Florence is probably your best bet. From there you'll take a train to Vernazza.
From Vernazza you'll take trains to Florence, and fly out of there back to Madrid.
Editing to add: as someone above is wondering, are you sure you didn't mean Milan instead of Madrid?

To cut to the bottom line: If I had tickets in and out of Madrid for an 8-day trip and did not have family or a business obligation waiting for me in Italy, I wouldn't be going to Italy. There are plenty of great things to be seen in Spain--more than enough right around Madrid to keep you busy for eight days.
