Train from Roma to Chiusi-Chianciano

At Chiusi you could take a bus to Montepulciano up the hill.
However I would rent a car during your Montepulciano stay.
You could rent a car at one of the companies walking distance from the Chiusi station (Avis-Budget, Europcar, Hertz are all there) or you could rent a car directly in Rome and drive from the start.

Like Roberto suggested, I would train to Chiusi and pick up a rental car there. Then drive to Montepulciano. I rented from Hertz on our last visit, and am doing the same on our next trip in September. The Hertz office is right across the street from the train.Just watch your times since the car agencies close from 12:30 to 3:00. So time your train accordingly. I recommend making reservation in advance. I do it from home before we travel.
