We Have hotel reservations in Positano in mid June. We have been reading that buses are often full with little to no room for bags. We are wondering what would be the best way to get to and from Positano from Sorrento with luggage? Any help would be much appreciated.

If you have a lot of luggage, then you might want to use a private car transfer. If you are coming from Naples, you might want to do it directly from Naples because the difference in price is not much whether you go from Sorrento or Naples. That way you save yourself a ride on the busy Circumvesuviana train.
Lisa,
The "best" way for those with a generous travel budget would be to arrange a private transfer with a firm such as Monetti Taxi (listed in the guidebook). I haven't priced it, but I believe the cost is quite reasonable.
Our luggage barely fit on the Circumvesuviana in late May. If you can afford an auto transfer, it will make your trip pleasanter all the way. Don't forget to ask your hotel about means of getting the luggage to the hotel if the car cannot drive immediately beside the hotel in Positano.
We're doing a private car transfer in April...from Naples train station to our Positano hotel and back to the train station a few days later. There's two in our party so the cost for each of us is a little over $100 round trip.
It is easy to ride the bus if you have a 21" suitcase ! We have done that several times.
If you want ease of travel, go to positanoshuttle.com or sorrentoshuttle.com - you can reserve a private shuttle or a shared for less cost.
Maybe do a shuttle as you arrive, and when you feel comfortable with the bus system, you can take the bus on the return.
