The cheapest optiond also depends on the number of people in your party. If it's just a single person or a couple the Leonardo train is the most affordable option. With 3 it depends if you would have to take a taxi from the train station to your hotel. If you can walk from there then again the Leonardo train is the way to go.
But if you would have to pay for a taxi on top of the train for 3 or if you are 4 or more then look into one of the shuttle services. We arranged for http://www.romelimousine.com/ last year and it was fast and affordable door-to-door service.

As the others have said, the Leonardo Express is probably your best option. Trains run frequently and the trip to Roma Termini is only about 30 minutes. You can buy tickets at FCO, but DON'T forget to validate your ticket before boarding the train.
Which area of Rome is your Hotel located? If it's close to Termini, you should be able to walk to the Hotel. If it's a bit farther, you could consider a Taxi (there are lots of them outside the front entrance of Termini).

Taking the Regional train, FM1 from FCO to Ostiense station, and following the signs to the Metro is the cheaper way to get into Rome. The FM1 is $4.50 Euros and the Metro is 1.00 Euro, making the trip into Rome from the airport $5.50, 1/2 the cost of Leos Express.
One small addition. If you do end up at the Termini train station we found it easy to use the city bus system to get to whereever we needed. Termini has a central bus staging site located right next to the train station. We found the buses cheap, easy to use, on time, safe and clean.
I believe that taxi fare is set at 40e during the daytime.If you have4 people its cheaper to take a taxi.
Most accommodations WILL arrange for a car to pick you up or drop you off at the airport. It is a flat fee of 40 Euro and is WORTH EVERY PENNY not to have to fight or figure out public transportation.
Sometimes least expensive and easiest (most convenient ?) are not compatible terms. The least expensive has been described but not sure you want to change trains by going from the train platform to the underground metro. And the same when you get to Termini. Not very convenient if you have more than one small bag. You also did not indicated where you staying, the number in your party. etc. -- all effect the quality of the answer.
We always stay within a couple of blocks of Termini just for the convenience of walking easily to the hotel. For that reason and because there are two of us, we find the Leonard Express as the most convenient and reasonable in price. But if you have a long taxi ride from Termini and there are two or more in your party, the fixed price cab at 40E could be more convenient and may be less experience.
I also look at the marginal cost. For us, our basic cost to Termini is 22E. On our last trip home from Rome we decided to use a shuttle for 30E. Hotel directly to airport and about an hour. I viewed that as only costing an extra 8 E but for that 8 E we were picked up at the hotel, did not have to walk to the station and equally important the long walk to LE track, buy a ticket, and the walk from the LE terminal to the airport check in. Returning we had an extra bag and some additional weight from the bottles we were carrying. I looked on that extra 8 E as a cheap price for the extra convenient of door to door service. There are other factors that you need to consider when determining if something is cheap and easy. Sometimes you have to pay for a little extra convenience.
