Would greatly appreciate any suggestions on how to get from Lucerne, Switzerland to the Lake Como area (specifically Varenna, Italy)
on a Sunday in late September.

Assuming you don't have a car, the fastest way is by train via LUZERN > LUGANO > MONZA > VARENNA-ESINO (one train change in Lugano and another change in Monza). A little over 5 hours in total. If you have a car, it's a bit over 3 hours.

Also Note that Trenitalia train schedule for late September is not up yet

You can look at same day of week now for an idea of schedule
It doesn’t change much and should be posted first week or 2 of June

You might also want to edit your subject line to provide a bit more specific info like
