Just take an official taxi that has a flat rate of 50 Euros (no tip) to the city. Just be sure to go to the official taxi rank and do not deal with anyone in the terminal offering taxi service. It might help if you have a 3x5 card with the hotel name and address written on it along with "50 Euros". You can get to the hotel cheaper, but you asked for the best way. After a long flight, and jet lag, and being on new ground, with luggage, you probably don't want to hassle with the train or shuttle buses.

Taxi is fixed rate 50 euros.
Walk outside terminal to official taxi rank, get in line.
Hand driver your address on a slip of paper.
Pay by credit card (confirm their machine is "working") or cash
No tip required.

The line at the official taxi location at FCO can be long and the wait can easily be 1 hour. We've found that hotels in Rome can sometimes arrange a private driver for about 10-15 euros more than the 50 euro fixed rate for a taxi from FCO into the city. And the private driver will be waiting for you right at the Arrivals door with your name on a sign. Just one more option.

I just wanted to add, even the authentic cabs will try to scam you. One wanted 150 Euro from airport to my hotel in Rome. So be careful.

We've always taken a taxi and never waited more than a few minutes.
We hand the driver our address and 50 euros cash when we get in the car. Eliminates scamming.

My guess is that topgun didn't have an official cab at the airport but rather one that approached him prior to the official cab starter/line. And I have never encountered a wait of more than ten minutes or so at the taxi stand. I am sure that on special holidays when traffic is heavier you could encounter a longer wait. But an hour wait is not routine.

I had a less than 5 minute wait at the official taxi rank. I paid the flat rate fee and there were no issues. It was a splurge for me to take a taxi, but I was glad that I did. I'm not sure I would have found my hotel the first time if I had tried to do this on public transport.

Five adults -- NO. Four adults and a small child or 3 adults and 2 small children - you will be OK.

My plane arrived at 11:30, so I had the not-so-brilliant idea to have the hotel book me a driver. My theory was that this would increase the likelihood that my room would be ready for check-in when I arrived. This was in October (2024). It was €65 plus a €5 tip. Well, I searched the throng of private drivers for my name to no avail. Turns out the driver was delayed or didn't count on me moving speedily through the airport. Anyway I had about a half hour wait but we texted each other so I wasn't worried. [I didn't have cellular service, used the airport WiFi.} And yes, the room was ready when I arrived at 1:30pm.
