Can one take a train from the cinque terra towns directly to florence or do you need to go to pisa centrale?

No, there are no direct trains from any of the C.T. villages (Monterosso, Vernazza, Corniglia, Manarola, Riomaggiore). Some change in Pisa, some in La Spezia, and some require changes in both. Explore your options on the Trenitalia website:
