My husband and I will be staying near Montevarchi for a wedding. My sister will be in Rome while we are there. We are eventually meeting in Montepulciano. Can you please recommend the best town/station to pick them up? We will have the car, pick them up where you recommend and then go on to Montepluciano.

They can take a train direct to Chiusi-Chianciano and you can pick them up there.
The advantage of picking them up in Chiusi is that Chiusi is a straight drive down the autostrada. You could also exit further north, then take local roads and meet them at the Montepulciano train station (very near the autostrada but there's no exit). The advantage of that option is that the drive to Montepulciano will be short. Either option would work well.
