4 of us arriving Venice 6/15 and must be in Trieste by eve of 6/19. Have Last Supper reservation 8:30 am on 6/19. What's the best route and method of transportation for 4? Want a day in Venice and the rest is open. Thanks.

Train from Venice to Milan and then train again from Milan to Trieste.

If you are landing in Venice, there is great bus service from two different companies from Venice-Marco Polo to the Piazzale Roma where you can catch the public vaporettos (water buses). Once you are ready to go to Milan, you go to the Ferrovia vaporetto stop (main train station) and take the train from Venice to Milan.
The trip will take 2.5hrs-3.0hrs with the first Eurostar train out of Venice at 5:32am and arriving at 8:25am. There is an earlier local train but it arrives at almost 9:00am. If you have an 8:30 Last Supper Reservation at 8:30, it doesn't look like you can get there early enough on the train. You might have to spend the night in Milan.
