Should I buy my train ticket now, or the day I travel from Manarola to Venice? I'm concerned about the CT train being behind schedule and missing a connection (looks like 3 changes) with only 10-15 minutes between connections.

Don't rely on a tight connection getting from Manarola to La Spezia. Those regional,trains often run late. I would wait to buy my train ticket to Venice.

The train from the CT towns to La Spezia runs often, you could just take an earlier train to give yourself lots of time to switch trains in La Spezia. I pre-booked all my long-distance travel, except from CT to La Spezia and vice versa. You can get cheaper ticket prices if you book enough in advance.

The trains in the Cinque Terre are reasonably punctual, so I wouldn't be too concerned about missing a connection, and 10-15 minutes is quite adequate.
The trains you'll be using on that route will likely be a combination of Regionale and high speed trains. It would help to have some idea on when you're travelling.
