The best and quickest way to get from Roma to Sestri Levante to Milano?

Trains (which ones?) or buses, or a combination? After a 4-day stay in Roma, we will visit friends in Sestri Levante for 3 days, then go to Milan for 2 days, where we'll fly home from.
