My partner and I are planning on spending September and October 2019 in Italy. We'd like to spend each month at a different location. We are interested in sight seeing, biking, cooking courses, and generally exploring. We are thinking of spending one month in Siena but would appreciate suggestions as to where to spend the second month. We won't have a car so someplace with easy access to trains would be ideal. Also if anyone has suggestions as where to stay or activities to do in either area I'd love to know.

While Siena is a very pretty city, I don't think it would be the ideal place to stay for a month without a car. It does have limited bus connections, but other than Florence, it's train connections can be rather convoluted.
A much better transportation hub would be Bologna.
As for another area, consider the Campania region. Naples, the Amalfi Coast, Capri, Ischia, Prodica. Pouzzoli, Baia. This area is so full of world heritage sites, it would take much more than a month to see them.
Both Bologna and Naples have all the things you're looking for. Both have easy access to Rome too.
There is no pace like Rome! Terrific train connections to everywhere and endless day trip potential.
Instead of Sienna, I would pick Florence for better transport.
If you like Siena, you might also want to consider Arezzo. It has a fun jousting festival in September, and good train connections to other towns. Another town that would be on my short list would be Verona (easy to enjoy the Dolomites, Venice, Ravenna, Milan, etc. by train).
In Verona, check out Veronality for cooking classes and bike tours in town or beyond. There’s a Verona Card if it’s your first time there to see the main sites.
Florence and Rome would be my choices.
