That is a very broad question and the answers are endless. Have you read a guidebook yet? When I was planning our first trip to Italy several years ago, I used Rick Steves' Italy Guidebook. I read the whole book, cover-to-cover, making notes about where I wanted to go. Of course everything sounded grand, so I had to tailor my expectations to the time allowed. We had 22 nights so were able to cover some ground. Looking back we covered too much, though. Now we go slower and enjoy more time in fewer places.

With only one week, you cannot go everywhere, but you can find history and great wine wherever you go. Start with reading a guidebook, then come back here with more specific questions.

By the way, with a week, you can really only do justice to one or two places, 3 nights in one place and 4 in another. The website/blog Italy Explained has a one week itinerary but I, for one, think it moves to fast/is too ambitious.

Spend 4 days in Rome and take a fast train (with reservations) up to Venice for the remainder of your time. If you had another week, you could take in Florence and Tuscany.

With only one week (which may or may not include travel there and back) I would stay in only one place. You can easily spend a week in Rome, Florence, or even Venice. Better to really get to know and enjoy one place than have memories of several places that are just blurs.

I would prefer you had 10 days, meaning you wouldn't have to choose between Florence & Venice as your 2nd city. Seven well organized days could see you do ... Rome (3nts) >> Florence (2 nts) >> Venice (2 nts)
An extra day in each would be preferable.
