What exactly is a single use room supplement? To me, it looks like an extra charge if there is just one person in a room. is this correct? And if so, why is that?!

Single room supplement means, you get a room alone. You don't have a roommate.
Yes, it is an extra charge for choosing not to have a room mate.
The why is, If a double hotel room costs 110 euros and there are 2 people in the room, it is 55 euros per person. Now, say in the same hotel a single room is 85 euros, that makes it 30 euros MORE per night for the single person. So, the single, who for what ever reason, chooses not to have a room mate has to pay the higher price as the room solo room costs more.

If you're traveling solo spend the extra money on a double room for single use. A single room will most likely have a single bed and be extremely tiny. A double room with have a bed big enough for two.
If there is high demand for rooms - and they only have double rooms available... if they use a room for a single and (if its a double bed) lose the opportunity to get a second person rate. If they have single-bed-rooms, then they usually shouldnt charge extra. Id check their web site first (as perhaps the only rooms left are doubles...) Also - if its a translation error - they may charge a premium for single-nite-stays if its on a weekend etc... vs staying 2 or 3 nites... just a thought. If you can call/email that may help. Have a back up hotel just incase!

If the hotel offers both single and double rooms, the supplement may be the extra charge over the single room rate for use of a double room by one person. In that case, the combined rate (single plus supplement) will probably be slightly below the standard double room rate. I suspect the slight discount equals their price for a second breakfast.
