Are you renting a car or getting around on public transportation? That could have an effect on your options.
But I would recommend Volterra and San Gimignano, Pisa and Lucca (a little further north), Montepulciano and Pienza (south of town), possibly Cortona and Arezzo, and even further afield to the southeast, Assisi.
The pairs I have listed above are close together, can be accessed by public transportation, and can be done in one day if you'd like to be ambitious and see more towns overall. I would limit it to just one of the pairs (two towns) per day at most, in order to at least get a feel for and experience each hilltown.
We will be in several rent cars. There are going to be 11 or 12 of us. We will have spent time in Florence before we go to Siena. As we leave, we are headed to Arezzo for the sale. Thx, B
Bart, not sure about when you want to rent the car and where you want to drive. As a caution, I hope that you have read the tons of info on this website about not driving in Florence. The whole historical area is a restricted travel zone except for the train station area. They get your license plate with computerized cameras. Arezzo has them also. Take a train or bus into Florence. Besides, you won't be able to find parking there anyway.
