My sisters in law and I are thinking of spending one night in San Marino after leaving Florence. Is this enough time to spend there and if so, what are some interesting things to do in San Marino. From the video I just watched and what I have read, it looks like a quaint area off the beaten path.

We were there yesterday in fact.
Spectacular place, no doubt. We were very pleasantly surprised. It it beautiful, make sure you walk the path between tower one and 2.
The rest are beautiful small streets packed with your typical leather shops, wine places, restaurants, etc.
But again, views are unreal.
You can take the cable car to go up if want. Just follow "Borgo maggiore" while following San Marino. The cable car is 4.50 euros round trip and the last one down is at 8pm.
If not you can go the regular road and go the different parkings. I think 4-5 hours there should be enough.
Never been but have researched it a fair amount. Paid for a hotel I never used there due to a last minute change of plans while driving.
I don't think it is a quaint place, gets tons of local day trippers/tourists from what I understand that go there for the duty free shopping.
I definitely think it is a great 1 night stay place. The towers are the main draw and the views from the town and the towers over the surrounding town and countryside. I think it's beautiful views are very under rated and hope to go someday.
I would want to stay overnight to be there for sunset personally and think after dark when all of the day trippers leave it is probably much more enjoyable and would at those times feel quaint.
Ideally you could go when the towers are open later than sundown, would have to research the closing hours and sunset times to figure that out and likely not possible most of the time.
Basically history, scenic towers and shopping is there, not much else so staying multiple nights is probably not great but plenty to keep you occupied for a one night stay if San Marino is not too far off track for you.

We really liked San Marino and we've stayed there twice. This was a place I've wanted to visit since I was nine years old. We first went there in 2014 for one night and liked it some much we returned in 2015 for three nights. I've read the negative comments by some posters, but we liked the place and would return again.
During both times there we did not see hordes of tourists and at night we had the streets to ourselves.
We stayed both times at the nice Hotel Ristorante Cesare and really enjoyed our experience. The second time we were there our room had a balcony, which provided STUNNING nighttime views.
Overall, the people were friendly, the restaurants were good, our hotel was great, and what's not to like.

I like it too. The only time there are hordes of tourists is in July and especially August, when the beach towns below it are taken over by the Russians and the Germans. Any other time it's kind of quiet, except for Sunday day trippers.
