I know ideally an overnight stay in Cinque Terre is preferable but do you think a day trip from San G to Cinque Terre is do-able? We would drive to La Spezia and take the train the rest of the way.

Your answer is contained in your question. Do-able,yes,but not preferable. If one day is all that you have to work with, give it a go!

If you already have a rental car, why not just drive to the Cinque Terre. Monterosso is (relatively) easy to get to, and there's a public parking lot on the beach.
