We will be renting a car out of Florence airport and travelling to Siena and ultimately Montepulciano. What would be best route to take? Should we save Siena for a day trip? We are in Montepulciano for a week.
Are you asking whether to stay there overnight or to see en route to Montepulciano? It's a fine city, well-worth a visit. A fun one at night too. But you could do it in a day trip, for sure. I would give it a full day though.

Consider driving down s222, the chianti road, and visiting some towns on the way to Montepulciano. Save Siena for a full day.
We are looking at what to do enroute. It will be a Sunday. We were thinking about doing the drive thru Chanti.
I would enjoy Route 222 for a Sunday and circle around Siena to get to where you will stay in Montepuciano. Come back to Siena Tuesday or later. Mondays in Italian towns are dull. On your drive south from Florence you'll pass through some very cool towns. Just plan on stopping and enjoying yourself along the way. One place I would stop for a glass of wine and scenery is the Mulino di Quercegrossa just north of Siena. You'll see a sign right on the 222.
You could also go to Siena from Florence spend a night and then rent a car. Florence to Siena is a easy 1 hr bus trip taking you to the Siena center.
