We are going on a tour that ends at the Vatican at 4:00pm and another tour that starts at 5:00pm from Piazza Navona. Is it realistic to think that we could make it to our 5:00pm tour on time? Google maps said that there is a bus that will get us from the Vatican to Piazza Navona in 21 minutes and I have also heard that Uber is banned in Rome? How reliable and accessible are Taxis at the Vatican? Any advice would be appreciated.

You'd have to run out of the Vatican and dodge a lot of people going every which way. (You know that there is a bag/security check at The Vatican? To avoid checking a bag, bring a small to medium size bag/backpack - around 14" should be good. - Anything big and bulky will need to be checked in their coat-check room. You will waste precious time going to retrieve your bags. No big umbrellas. Water bottles were "okay" when I was there, but they had to be put away in bag.)
No uber, correct. I can't attest to the official Roma cabs - how busy or how fast they can get you to Navona.

In case you are not aware - No bus tickets are sold on the bus. No change is made by the driver. Get your bus tickets (ahead of your Vatican tour) at a Tabacchi shop (T), a little market or some gift shops.
Validate the tickets when you get on the bus. Transit Inspectors will stop a bus at random, board and check everyone's ticket or pass. If you don't have a valid, stamped ticket, you could or would be fined a lot of Euro. Bus tickets are good for 100 minutes (and a transfer) of travel.
When riding Bus #64, keep your valuables close - in front of you if possible. No money, Passport, or phone in back or side pockets. Don't give anyone temptation or cause.
P.S. An additional thought: It happened to me. If, by chance, the machines are invalid, a Transit Inspector showed/told me what to do if that happens again - On the back of your ticket, write down the stop you got on, time you boarded and bus # (i.e. 64); also, he said it would be helpful if the Bus ID # - located on the rear of the bus - could be noted so they can analyze the non working machines. I did not get fined because it was proven the machines were broken. It's good to heed the recommendations of authority.
If you aren't exhausted, you can walk from St. Peter's to Piazza Navona in about 20 minutes. Less hassle.
I'd plan to walk (or run) so you are not subject to traffic delays etc.
But I have to say- we have done a lot of diff tours in a lot of places and they almost always run over- so you may not actually really be DONE at 4 exactly.
Does it actually end at Vatican Museum or at St Peter's?
Is this the only spot in your itinerary for the tour that starts in P Navona? Do you really want to go from one guided tour right into another?
Voting with Laurel and Christine: walk it. It's really not that far but it would be helpful to know exactly where your Vatican tour ends.
Thank you for the replies. We are only in Rome for 1 day and want to get the most out of the day because I don't know if we'll ever be back. I know it will be a long day but the second tour is a food tour so we'll be sitting a lot of the time so that will help.
Ah- well we love food tours so I see your point!
But you will still be doing a lot of walking not just sitting.
Anyway- I might contact the food tour company and ask them where the first "tasting stop" is- just in case you are late- you can always catch up to the group there.

Not a bad walk at all. Just make sure you have a good map, it’s a straight shot across the river but still easy to get lost in those alleys
I would cross the river then stay on the main drag Corso Vittorio Emanuele
Also mentioned above- don’t bring any bag that you would have to check at Vatican or you’ll have to go back to entrance to retrieve
