Will be taking the SENA bus from Rome to Siena tomorrow. They offer purchase online but apparently NOT to US citizens(!)
I'm (nervously) wondering if it is absolutely certain that one can step up to the counter some time prior to departure and simply purchase a ticket at the Tiburtina Station counter?

You should have no problem, get to the station early enough to cope with possible lines at the ticket window. Worst case is you would buy a ticket for the later bus.
I am not sure about this bus - never taken it. However, does is specifically say it won't sell online to US Citizens? I ask because on the ALSA bus in Spain, you can't use a US credit card but can use PayPal. If your problem is with a card, try paypal. If it's more fundamental than that, I'd do as the previous poster said and arrive early.
They certainly will sell tickets up to departure, as long as the bus is not full, which I would not be too worried about.
