Should we buy our tickets online before we leave for Italy? We will be taking the bus to Siena from Rome on June 20, Friday, or should we buy them when we arrive in Rome June 15.

In another post you wrote you and your family will have a car in the Siena area. Why do you need bus tickets then? If you intend to rent a car why don't you rent your car in Rome and drive to Siena? There are a bunch of car rental companies at or near the Tiburtina station where the Siena bus departs from. It makes no sense to go to the trouble to take a bus to Siena to pick up the rental car in Siena.

Never considered renting a car in Rome. Just heard too many horror stories about having a car in Rome. Will look into it.

You don't rent it while in Rome, you rent it on the day you leave Rome. From the Tiburtina station you can easily access the autostrada (freeway).

The Tiburtina station is outside the ZTL zones in Rome, no worries about that.
