What is the quickest and best way to get from Rome to Siena?
"Best" is very subjective. Can you be more specific?
Quickest is probably by train. It will take about 3 hours with a transfer in either Florence or Chiusi. There may be a bus but I don't have any information on that.

As with your other trips, I'd recommend travel by train (although you could also go by Bus). One of the quickest rail journeys is a departure from Roma Termini at 08:58, arriving Siena at 11:57 (time 2H:59M, one change at Chiusi-Chianciano Terme). When you arrive in Siena, you'll have to take a Taxi or Bus up the hill to the town.
I disagree with advice to catch the train. The bus takes you around the same amount of time as the train and has the advantage of dropping you close to the centre of Siena, not at the station which will probably require a trip to your hotel by taxi. You also dont have to worry about changing trains.
Buses go from Tiburtina station. Marginally further from the centre of Rome than Termini station, although this depends on where you are staying.

I agree with mph, the bus is easy, convenient and drops you off right near the city center.

The bus will get you into Siena faster and you don't have to haul bags up and down stairs.
I agree with the bus. My problem is trying to purchase tickets through the SENA site. Because I am purchasing from outside Italy I think I will have to use the SISAL option. Has anyone ever used the SISAL pick up option? If so, what is involved in this option and did it work for you?
