Train. 1 hr 35 mins, city center to city center. Go here to see the schedules: http://reiseauskunft.bahn.de/bin/query.exe/en
On the site, date isn't important, from: Rome Termini, to: Florence SMN. Enter an appropriate time to see the schedules for the time of day you want. You can buy tickets when you get to Rome.

Galina, For information on buying tickets from the Ticket Kiosks at Roma Termini, have a look at This Excellent Website. Scroll down to "Buying Train Tickets". There are staffed ticket windows at Termini also, however you'll probably find there's a perpetual and often huge queue at the ticket windows. A credit card is normally required to purchase tickets from the Kiosks (at least that's been my experience there). You'll be arriving at Firenze Santa Maria Novella, which is the main station in Florence. Depending on the location of your Hotel, you may be able to walk from the station. Are you travelling directly from the airport to Florence? Cheers!

The standard 2nd class fare for high-speed EuroStar Italia trains from Rome to Florence is €45.00. However, if you book your tickets now on the Trenitalia site, you could get a cheap Mini fare. They can be as low as €22.00.

The trains are crazy easy. Use the little self serve kiosks in the stations and book second class base. Don't wait in the ridiculously long lines. We just returned yesterday and hauled 8 people (4 kids, 4 adults) and all their luggage this way for 2 weeks. If only the US could pull this off.
