If you use Trenitalia: ROMA TERMINI
If you use Italo: ROMA TIBURTINA

Jim, some may argue about which is the best train from Rome to Florence - Trenitalia or Italo but Rome-Termini has about 55 high-speed trains a day departing for Florence. Most are 1:35 non-stops.
