Rome, Cinque Terre, Florence and Venice

Would you travel from Rome to Cinque Terre or go Rome to Florence first and then Cinque Terre ending in Venice?
So Rome, Cinque, Florence, Venice or Rome, Florence, Cinque and Venice?

How many days for your trip? You should do at least 6 days in Rome, 3 in Florence and 3 in Venice.

If you do go to CT, I would suggest going there after Florence.

Depends on what month, how much time you have, and what you plan to do there--it's going to require an excessive time on a train IMO.

My wife and I did this itinerary in September of 2021. We chose to go from Rome to Venice because there is high speed train service with limited stops for the longest leg of the trip. We then went to Cinque Terre and Florence . The long ride to Venice was a welcome rest after five days of walking all over Rome. We also like to put some relaxation in the middle of our itinerary and Cinque Terre is perfect for that.
The rail system in Italy is really good for the places you are going. The frequency and reliability of trains gives you the flexibility to visit these places in whatever order feels right for your travel style.

Travel Scamp, the OP is flying out of Venice so Rome>Venice>CT> Florence doesn't work.

I'd probably do Rome>CT> Florence> Venice.
You can do a shorter journey with fewer changes from Roma Termini> CT (I'm using Monterosso)
9:57 AM, 4h 41min, 1 change (at La Spezia Centrale, which is a gateway to the CT)

Shortest morning journey from CT>Venezia S. Lucia
09:13 AM, 5h 21min, 2 changes (at Pisa and Florence)

There is high-speed rail from Florence to Venice.

What Kathy said. Most logical to avoid backtracking is Rome, CT, Florence, Venice. Then CT to Florence (station Firenze SMN) is 3.5 hours, Florence to Venice (station Venezia S Lucia) is 2+ hours. (I think Kathy may have mentioned CT to Venice by mistake, as you would be stopping in Florence). Have a great trip!

Sorry Bob - what I wrote was probably confusing!
I used the CT>Venice time as a comparison against the Rome>CT time to show which choice was shorter and took fewer changes, thus the Rome> CT> Florence> Venice recommendation versus Rome>Florence> CT> Venice.

Does that make sense now?
