Rome-Assisi-Cortona-Siena-Santa Margherita

After arrival in Rome, I head straight to Assisi for 2 days, then to Cortona for 2 days, then to Siena for 3 days and then to SM for 2 days - then back to Rome for one day before flying back. Should I do trains or buses? Thanks for any help.

Great places to see ? I would do train because it sounds like your trip is short so your time is precious. We have stayed in SM and stayed in Spello 10 min from Assisi

I agree with David. All of the towns you plan to visit have convenient train service, so that's the way to go. You'll need to take short bus trips from Assisi, Cortona and Siena train stations into the towns. Trips take only a few minutes and cost about 1 euro. Train station in Santa Margherita Ligure is only a short flat walk from the town center.
