We will be arriving at Fiumicino--wanting ,then, to get into the city. There will be two of us. What is the "best and easiest" way to get into the city center?
2 ways depending on your preferences, budget, and amount of luggage:
1) Take a cab from the airport to the door of your hotel. This is the easiest option, especially if you have a number of bags. Depending on where your hotel is, it should cost you a flat 40 euros to get there. Be sure to get only into an official marked taxi and don't let the driver pull a scam on you. Tipping is at your discretion, if the driver has gone the extra mile, helped with bags, not scared the life out of you with his driving, etc.
2) A cheaper alternative is to take the Leonardo Express train from the airport to Termini station. This costs 11 euro I believe. Follow the signs from the baggage claim to the train tracks (there's only 1 so you can't get on the wrong one). Purchase your tickets from the shop there or from the ticket machines which accept credit cards to save you the hassle of needing euros right away. Validate your ticket in the yellow box before boarding.
The train ride takes about 30 minutes and ends at track 24 of Termini station. From there you can walk, ride the metro, or take a taxi to your final destination (depending on where that is).
Option 3: Arrange for a driver to pick you up, e.g. Rome Shuttle Limousine for 35 Euros.

I think the best and easiest for two people is a limo. You can reserve one for 35 or 40 euro about the same as the train to termini and a taxi to your hotel. I have used Rome Shuttle limousine http://romeshuttlelimousine.com/ with good results but there are others.
Take the Leonardo Express Train into Termini for 11 Euros it takes 30 minutes and is very economical.
The Rome Shuttle was great for me! I was too tired to take the train, and having someone to pick me up for a flat fee was good. The driver barely spoke English, but that was alright! I speak Italian anyway, but even if you don't, he'll know where your hotel is.
Have a wonderful trip! I LOVED Rome!! But the traffic I didn't love so much! A taxi could get very expensive.
Beware of taxis from airport to Rome they try and rip travelers off. If you choose taxi get an cost in writing that incl all charges in advance! suitcases are xtra.
I would suggest Rome Cabs. it is a transfer and sightseeing service that is totally reliable and very dependable. We are charged 45 euros to take a new mercedes (not that it matters) from FCO to our hotel. Stefano and his crew are very dependable and give insight to various parts of the city as you drive in. Having used this service the last 5 trips i couldnt be more please. On our last trip, we had a CAR that was supposed to pick us up at 345 in order to get us to FCO for a 6 am flight. on this trip we did a whole lot of shopping and with my mom and our bags numbered 4 large checked bags. i knew there wasnt going to be enough room in the car so i called Rome Cabs to alert them at 9 pm and they were still able to accomodate us with a mini van type and at the same rate still. The express train from FCO is also a great way to get in and we have taken this route as well. if you are more centrally located to Termini, i wouldnt have an issue taking the train in but if you are closer to the historic area of rome, then i would deifnately arrange a transfer car.
As an alternative to the Leonardo Express, you can take the "other" train from the airport for half the cost and get off at one of the other stations and transfer to a tram or bus (or Cab)from there. If you are staying in the Trastevere, this would be easier than going to Termini then backtracking.
