I am traveling via rental car to Venice, my end destination. I’d like to know the best return location. Train station or airport and from there the best way to get to our hotel on the grand canal.
I would avoid the Hertz location at St. Mark's Square. Nothing but bad reports.

There is no road access to the railway station in Venice.
Piazle Roma is the clost you can get with wheeled transport.
I would suggest de-hiring the car at Marco Polo airport, and taking a bus to Venice. Bus transit time is about 20 minutes.
The bus will drop you at P.Roma, from there take a vaporetto (water bus) down the Grand Canal.
No need to spend extra time and money to go to the faraway airport.
Return the car at Piazzale Roma. From there take the vaporetto to your hotel.

To get there you will cross the Ponte della Libertà (Liberty Bridge) which connects the mainland to the old city in the middle of the lagoon.

As you can see in the image below, although the roadway is conducive to speed, don’t!
There are plenty of speed cameras on the bridge, and you will get fined if you exceed the speed limit by more than 5km/h (3mph) above the speed limit.

I agree with Roberto: there is nothing particularly hard about driving over the bridge to the parking lots in Venice, and that's what I would do!
The warning re. speed cameras applies throughout Italy, by the way.
