Where is the best spot to pick up a rental car when we leave Venice? We will be dropping the car off in Rome and will have a pickup to our apartment there. Is the main airport the easiest drop off point?

There are several agencies that have offices in Piazzale Roma in Venice. So no need to go to the airport if you don't want to.
Be sure to check hours though, I believe that a lot in P.le Roma have reduced hours over the weekend.

There also a Hetz place on Santa Croce which is right near where Ron mentioned.
