I an traveling to Italy in May and need rent a car in Florence, travel to Umbria, and drop it off in Rome for my flight home. Do rental car companies allow you to rent in one city and drop off in another.
In general, they do. There may be an extra fee (probably modest) for doing that. (It's dropping off a car in a different country that often comes with a very large price tag.) You could use one of the consolidators (AutoEurope, Gemut, Kemwel) to determine your best options.

We have rented in Italy with different pickup and drop off cities two times, both with Hertz. There was no extra charge as far as we could tell.
We have our vehicle booked with Discover, picking up at Florence airport and dropping off 10-days later in Venice. The cost of "One-way rental" is €86.38.
