Hello We are renting a car in Sondrio and driving to Ortisei for a few days then onto Venice. I'm looking for recommendations as to where to drop the rental? Thanks

Yes it matters which rental agency.
If you are spending time in Venice you do not need or want to go to the airport to drop car so it would be Piazzale Roma or Venezia Mestre.
From Mestre you would take the quick train into Venice proper.
From P Roma you can walk or take a vaporetto to wherever you are staying
The choice should be dictated by where the hotel in Venice is.
If you plan to stay at the Venice airport to catch a flight the next morning, then return it at the airport.
If you plan to stay in Mestre, then return it in Mestre.
If you plan to stay in the historical center of Venice, in the middle of the lagoon, then it makes sense to return it at Piazzale Roma.
Most major companies have offices at all of the above locations, although I think Avis-Budget has closed the Piazzale Roma location.
Be aware that on Saturday afternoon and Sunday all day, only the airport locations may be open.
Yes, I should have given more information. From what I can tell, getting an automatic transmission in Sondrio is from Lacauto.
We will be staying in Venice in the San Marco area for 4 days and then taking a train to next destination. This is a big belated birthday trip for my husband and he wanted to take the Bernina Express from Chur into Italy and then have a stop in the Dolomites. I think we are trying to do too much but... We have been to Italy several times and are revisiting a few places we have been already but the Dolomites is new for us. Thank you.
It looks like Lacauto is in partnership with Enterprise so you would have to check with the agency to see where you can drop the car.

Just checking with them they have automatics in Sondrio at Avis and Budget with drop off at VCE airport or Mestre.
Since you are staying in Venice a Mestre drop off makes the most sense.
As Roberto mentions above- be sure to check the open hours at the Mestre location
Make sure all drivers have an IDP- required by law.
