Family of 5 adults looking for lodging recommendations for the following cities/areas: Venice(3 nights), Florence(3 nights), Cinque Terre(1 night) & Rome(4 nights) for trip in early June 2018. (Will be traveling by train, not renting a car).
We have twice stayed as a family of 5 in Rome at the Hotel Madrid Roma. They have a suite that has 2 different rooms. The hotel is near the Spanish Steps which is a convenient location for walking to most of the main attractions.
Try AirBnB or booking.com. There are many apartments listed. Book fast, especially in Cinque Terre. If you find something, grab it!

I like to stay within walking distance of the train stations.
Much easier to get in and out of each place.

On a tangent to your question, you may want to reconsider the one-nighter in CT
Depending on the collective interests, cut Florence to 2 nights and do 2 nights in CT, or skip CT and add another night to Rome.
Into CT one day and out the next is a lot of bother for very little time actually there.
