We will be traveling from Rome to Florence in early October, and plan on taking a trail (hopefully).
After our Florence stay, we are hoping to rent a car in Florence and drive to La Chiusa in Tuscany and stay for a couple of nights.
But - after that, we will need to get back to Rome airport for our Hilton night before heading home. Wonderly if we should take a chance and drive the car back to Rome airport and drop it off there? Or, go back to Florence, drop it off there and take a train to the Hilton?
This is our first time in Italy, so appreciate recommendations!

There could be many places called La Chiusa--could you share the specific location?
This is really just a personal preference unless it is more than half way back to Rome. You may prefer the scenic drive over any inconvenience. But do be mindful of leaving luggage unattended in the trunk.
In Tuscany, we will be staying at a resort called La Chiusa.
Can you share a link to the resort?
Tuscany is a very large area

If you are going to the airport I would just drive there.
Your other option is Chiusi.
I would not drive back to Florence- that'll be a more difficult return

Think we’ll just drive directly back to The Rome airport. Just in case there’s a train strike, we’ll be covered. Thinking if we use Google maps, everything should be fine if we Return the car that way.

Return it at Fiumicino airport. The rental car return is in one of the multistory parking towers across from T3 (tower A,B,C,D) Which one depends on the rental company, but I think they are all in either tower A or B. Hertz is in tower A 1st floor. So follow directions to Rental car return, but then pay close attention to the signs of the rental companies so that you go to the right one. Don’t return the car in Florence. Too much hassle, besides the long back tracking.
Once you return the car the walk to the Hilton is 5 minutes on the same walkway which connects the terminals with both rental parking and the Hilton.
To drive in Italy use Waze. I use Google Maps in the US but Waze is best to warn you about speed trap cameras. There are a ton in Tuscany and most Italians use Waze so it s constantly updated. If you have never used Waze before (Waze is owned by Google), try it before going, so you become familiar with the icons etc., in particular the ones below:

For your autostrada trip to the airport you can use either since on the autostrada there are no speed traps, only the Safety Tutor System, which calculates the average. Plus the speed limit on the autostrada is very high (130 km/), so I is unlikely you will go above.
On the way to the airport this is the place where you need t refuel your car. It is the Area Di Servizio PISANA INTERNA. Set your navigator to this location first or remember the name of the Area di Servizio above. It is the last one before getting on the freeway to the airport. I always refuel here before returning at FCO (including last week).

If we rent the car in Florence and drive it back to Rome airport, do you know if there’s a drop charge?
We always drop off at diff location and have never had a drop off fee

I think Hertz charged me 15€ last time I dropped off at a different location. Europcar never did.
If you are concerned about finding your rental return at FCO you can use google street view and “drive” the route into the airport. The resolution is good enough that you can read the signs and see exactly where you will be going. I like to have an idea what to expect.

Just as a note, I booked through AutoEurope with Europecar as recommended many times here by Roberto and Christine (thank you both as I have read many of your posts!), picking up in Florence at SMN in October and returning to FCO a week later. The listing and my voucher said it will be a 135 EUR charge for dropping off somewhere else, to be paid locally. I guess we will see if I get charged, but I am expecting I will have to pay it. Autoeurope clearly listed this in the rates.
