recomendations on 2 days in tuscany

we're flying into Florence in the morning and will have most of two days to ourselves before heading to Rome to connect with our Heart of Italy Tour. We'd like to head into Tuscany until then, hit some beautiful scenery, wine tasting and food tasting and especially spending time in Siena. I'm looking for recommendations for:
1. guidance on auto rental. Our plans right now do not include going into Florence if we don't have to. We will want to return our car before we head to Rome.
2. any advice on getting back to the train to Rome or the train experience.
3. recommendations on countryside route. We want to see San Gimignano and Siena but not sure of which others. We'll hit Volterra on the tour so not trying to fit that in here.

4. lodging for 2 nights;

5. any outstanding food experiences that you want to share!

Picking up a car at Florence airport is a fine plan if you're heading to a smaller town, but if you're overnighting in Siena first, you can get there by bus and pick up a car upon departure. You can return the car to any other spot in Italy that the rental company offers, such as Orvieto (a lovely hill town on the main rail line), Orte, or Rome airport. If you want a car drop closer to Rome, search above for earlier discussions on this forum.
