Reading Train Schedule

On a trip from Milan to Venice the trenitalia site reflects all stations for both Milan and Venice. On the detailed info for Venice there is arrival in Mestre with no departure time and then listed below arrival in Venice S Lucia. Does that mean the train stops at both stations? Confusing as there is no departure time from Mestre

Yes, the train stops in both stations. Mestre is just a quick stop. They generally only show arrival/departure times in major cities where they might linger longer.

If you purchase tickets to Venice you'll want to purchase them to Venice S. Lucia station. Most trains stop in Mestre for a few minutes but continue on to S. L. Donna

Italian trains are confusing. you will get used to it. Best advice is (once you get on the right train), locate the conductor (tends to be in the middle during the first bit of the trip) and ask him any questions you may have. They may seem a bit stern and serious but they are very helpful and understanding and (I believe) required to speak a fair amount of English.

Don't wait to ask the conductor. If you are on the wrong train, that is too late. Years ago after getting on the wrong train going in the wrong direction we now always ask other people standing on the platform if this train is going to _____. Even if they do not speak English, just point to your ticket. They will be helpful. Your ticket will show the train number, the car no, and seat no. Only the regional trains do not have reservations. Use the train number when checking the arrival/departure board. After the first train, you will find that it is very easy and simple to use.
