Although I have not been to Ponza, I have researched it and it is easy, although it would be a very long day and I would spend the night. The train from Rome Termini to Formia-Gaeta takes an hour to an hour and a half, and the ferry from Formia to Ponza takes from one hour and forty minutes to two and a half hours (slow boat, fast boat).

What is attracting you to Ponza? It's a summer-oriented destination and you may find that most tourist-oriented businesses will be shuttered by October. There are many easy and lovely daytrips from Rome including Ostia Antica, Orvieto and Tivoli/Hadrian's Villa. Ron in Rome has excellent info on those here: http://www.roninrome.com/sites-attractions/outside-rome
