We are travelling to Italy next week and we will arriving into Venice airport. Is it better to buy a sim card at the airport or wait until we get into town? We will be taking the water bus to our our hotel.

None of the mobile phone providers in Italy have a store at the VCE airport, according to their respective websites.
CAPI Electronics, a European retailer with various stores inside European airports just opened a store at VCE airport last month, but I'm not sure they are authorized dealers for mobile phone SIM cards with any of the main Italian companies (TIM, Vodafone, Wind3).
You probably have no choice but wait until you get to town.

I would think finding in-town locations in that particular city could be a challenge. Ask your hotel front desk before you arrive for the nearest phone store.
You can find online the closest stores to your hotel:

Enter under address (indirizzo) your hotel address or just Venice
Under 'tipologia negozio' checkmark both options (TIM store or authorized dealer)
Do the same for Vodafone, but on the upper right side of the map under 'tipo di negozio' (type of store) select also the authorized dealers to get more hits.

There is one Vodafone store near Rialto Bridge, but there are several authorized dealers. This is the Vodafone store:
