I will be driving to and from Venice, staying over only 1 night. Where is the best place to park a car?
The only place is Piazzale Roma, at the base of the causeway from the mainland.
Not to sound like a broken record, but the way to visit Venice is NOT with a rental car and NOT for one night. Those 2 variables will quite possibly cause you to have a miserable time in Venice. Why?
1) Long lines of traffic trying to get into a packed car parking garage.
2) Dragging your luggage from the garage to Vaporetto
3) Probably a PACKED Vaporetto at that
4) You'll be hot, bothered and tired by the time you drag your luggage around trying to find your hotel
5) You'll wake up the very next morning and have to do the exact same itinerary in reverse with the exact same conditions.
Other than that have a great time in Venice.
Nobody uses Tronchetto anymore? Cheaper than P Roma, less traffic, has its own Vaporetto stops, as does P Roma.
By the way, it has been my experience that vaporettos, especially the number 1 which starts there, are considerably less crowded at P Roma than just one stop further down the Grand Canal at Ferrovia (the train station). Many hotels, al the ones I now regularly use, have routes which cross no bridges to the vaporetto stop.
You can also park in Mestre. There is a parking garage across from the station. You can leave the car there and proceed to Santa Lucia station by train. Mestre garage is a bit cheaper than Piazzale Roma.

Agree with Nigel and not Tim. Crowding is worse at the station in my opinion. As for driving, sometimes there is no sensible alternative, Tim. If I have lease car for two months, what else am I going to do but drive I'd my itinerary includes Venice.
I assume you are more than aware of the pros and cos of a car, and have made your own, informed decision that it works best for you (it did for me). I used Tronchetto this past July, with none of the unpleasantness Tim describes. It was reasonably easy to find a spot to park, they have a secure luggage storage (in the manned office where the payment clerks are) and it's right across from a Vaporetto stop. The Vaporetto was uncrowded (available seats with good views) to start, but became more so as it went down the Canal. The secure luggage drop allowed us to lighten down to a single small bag so "dragging" luggage wasn't a problem (and if we took our bigger bags, they have wheels!).
I do agree with Tim that you should have a great time.
