Overnight in Umbria near Assisi

We'll be in Rome the first 10 days of October (renting an apartment with another couple). We'd like to do an overnight trip to visit Assisi and then maybe tour a winery in Umbria.

Questions: Where should we stay? Should we rent a car in Rome and drive to the region? Should we take a train to the region and rent a car there? Can anyone suggest a winery in the area?

Why not stay in Assisi?

Montefalco is a great spot for visiting a winery, although I don't know of a specific tour, they do exist. Also, there is a terrific restaurant and wine store in Spello called Enoteca Properzio that has wine tastings.

We loved Spello! Spent 4 nights there making it our base for Umbria. One wonderful day we let Mark from Gusto Wine Tours take us around. We visited 3 great small wineries that you most likely would not find on your own, and had a lunch to remember at an agriturismo he knows. No car required: take the regionale train and spend a couple of nights enjoying the area. Ask Mark for ideas.
