No personal experience.

Looks to run only 4 days per week. Departs Palermo at 2AM and arrives Civitavecchia 13 hrs later.

The port is 50 miles from Rome so you still have 2 hrs more to get to Central Rome

You sure you want to spend that much time in transit versus flying?

Another possibility, which is non flying but incorporates a ferry (20 minute crossing) is the Palermo to Rome train which uses the train ferry across the Strait of Messina.
There are 4 trains daily-
0650 arrive 1834; 1000 arrive 2138; 1848 arrive 0718 and 2110 arrive 0951.

The night trains have seats, couchettes and sleeper cabins. Fares start at 20 Euros, or 30 Euros for 1st class by day or about 150 Euro for 2 people in a Premium sleeper cabin. Or 80 Euro in a solo 1st class cabin overnight.

There are also ferries from Palermo to Naples, then train to Rome.

Back in 2015 I had an utterly miserable, sleepless night traveling in the opposite direction on that night train in a couchette car. The carriage jerked constantly from side to side. I assumed the rails were in really bad shape, but someone else here reported the trip was OK in a sleeper car, so maybe the issue was my couchette carriage.

Haven't taken the overnight ferry from Palermo to Rome, but if this helps...

We did the overnight ferry crossing from Naples To Palermo aboard the Rafaele Rubattino. The cabins are basic but comfortable, the sailing was smooth, and we slept well. We saw a sunset in front of Mt Vesuvius and then a sunrise approaching Palermo. Here are a few pictures of the ship and cabin...
