Ride the funicular up then hop a bus for the next leg which will drop you right in front of the Duomo
Definitely! Just realize that when the train leaves Rome, it is on a very isolated track far outside the terminal, so allow enough time to get there. And I recommend walking away from the main square (duomo) to get away from the crowds. It's a lovely change from Rome.
Orvieto was my favorite part of my Venice-Orvieto-Rome trip last year. I recommend the underground tour (1 hour long, I recall) and St. Patrick's well was pretty cool too. Can you stay a night? It's a really pretty town to walk through at night.
