My husband and I are taking the train from Orvieto to Florence in June for one night. We are then going to Venice for two nights. What are the must dos/see in such a short amount of time?

In Florence I would go to the Duomo Museum.
In Venice - St. Mark’s church, gondola ride, Grand Canal by vaporetto

With 1 night in Florence and 2 nights in Venice, you'll really only have an afternoon and evening for Florence and a day and a half for Venice. The suggestions above for Venice are good and you should also have time to visit the Ducal Palace, but in Florence I suggest you pick either 1 major or 2 lesser sights to visit in the afternoon and then spend the rest of your day walking around to always-open sights like the Ponte Vecchio, having a nice dinner and just enjoying the city. Since you haven't mentioned your interests, the suggestion to get a guidebook and scan it for the sights of greatest interest to you is sound advice.
